Position Summary

We are looking for a motivated biologist to join our Platform Biology team as a Research Scientist. This position will be dedicated to designing and conducting experiments involving cultured cells or isolated proteins and applying multiple methodologies to develop protein degrader small molecules.

This is a primarily bench scientist position, requires regular presence at our headquarters in New Haven, CT, and reports to the Research Investigator in our Platform Biology group.

Principal Responsibilities

Key responsibilities of this role include, but are not limited to:

  • Design and development of DNA constructs, cell lines, and cell-based assays.
    • Manage DNA construct production by CROs.
    • Conduct in-house and manage external cell line editing (CRISPR/Cas9).
    • Cell clone generation.
    • Application of lentiviral and/or AAV systems.
  • Development and execution of cell-based assays to query targeted protein degradation.
    • Antibody-based techniques: western blots, in-cell westerns, ELISA, flow cytometry, etc.
    • Luminescence- and -fluorescence-based assays: reporter assays, NanoBRET, etc.
    • Cell viability/proliferation assays.
    • Cellular target engagement assays: NanoBRET, CETSA, etc.
  • Design and execution of degrader mechanism of action studies.
    • RNAi experiments and/or use of edited cell lines.
    • Compound dose-response curve generation: IC50, DC50, Dmax, etc.
    • Application of numerous available chemical tools: UPS inhibitors, probes, etc.
    • Work with Arvinas chemists to design novel tool compounds.
    • Conduct cell-free biochemical assays (e.g., ubiquitination).
    • Protein immunoprecipitations and pulldowns.
    • Partner with internal and external proteomics collaborators.
  • Work with multiple teams and team members in complex scheduling of work and generation of data in support of drug discovery and development programs.
  • Troubleshoot through technical and scientific challenges in order to deliver critical data.
  • Analyze multiple types of data, applying a strong background in quantitative cell biological sciences.
  • Apply excellent organizational and time management skills.
  • Maintain clear experimental records and communicate well with supervisor and team members.

Qualifications

  • Motivated, creative, and productive problem solver with excellent communication skills who works effectively in a team setting.
  • Proficiency in aseptic mammalian cell/tissue culture is a must.
  • Extensive experience in molecular biology techniques: plasmid and cell line engineering
    • Lentiviral system experience.
    • Experience in generating CRISPR/Cas9-mediated KO or KI cell lines is a plus.
  • Extensive experience measuring protein levels: western blot, ELISA, reporter assays, etc.
  • Experience in execution of medium- to high- throughput (96/384-well) plate-based assays: examples include ELISA, TR-FRET, quantitative western, luminescence- and/or fluorescence-based assays.
  • Strength in assay development and execution using multiple methodologies.
    • Experience in utilizing compounds and molecular target manipulation (e.g., RNAi, CRISPR/Cas9) to dissect target function and dependency.
    • Biochemical assay development: cell-free ubiquitination, immunoprecipitations, pulldowns, stained gels, FRET assays, etc.
  • Expertise in cell biology, molecular biology, and biochemistry.
    • Knowledge and understanding of the ubiquitin-proteasome system is a plus.
    • Knowledge of drug discovery approaches is a plus.
  • Organized, goal-oriented, and timeline-conscious.
  • Other preferred skills and knowledge.
    • Experimental documentation/lab notebook.
    • Data presentation to diverse audiences.
    • Experience with Microsoft Office suite (Word, Excel, PowerPoint), GraphPad PRISM, Pymol (or similar), Benchling, and other industry-relevant software.
  • A strong work ethic and willingness for continued learning.

Education

  • Ph.D. in Biology or related field with 0-6 years postdoctoral experience in either academia or industry.
