Position Overview:

As a postdoctoral fellow in Merck Research Labs Chemical Biology group based in West Point, PA, you will work with a highly collaborative team of scientists to design and utilize novel methods for genetic code expansion to study intracellular protein interactions and dynamics.  This research will enable understanding the molecular mechanism of action for chemical probes with compelling phenotypes and provide insights on therapeutic strategies for pathways which have proven challenging to modulate clinically.

Responsibilities include but are not limited to:
  • Develop strategies and experiments based on genetic code expansion to address knowledge gaps in the understanding of disease biology
  • Design mutant proteins incorporating unnatural amino acids based on structural and functional data
  • Design constructs for co-expression of mutant proteins and orthogonal tRNA/aminoacyl-tRNA synthetase pairs; optimize expression efficiency
  • Design and conduct live cell imaging studies using a variety of techniques including but not limited to in-cell “click” reactions and confocal microscopy
  • Design and conduct cell-based interaction studies including photocrosslinking of proteins containing reactive unnatural amino acids, immunoprecipitation, digestion, protein mass spectrometry, and data analysis
  • Collaborate with Merck and external scientists including Chemical Biology, Structural Chemistry, Target Protein Design, Neuroscience, Pharmacology
  • Data interpretation, manuscript preparation, publication and presentation of results
  • Contribute to a safe and compliant laboratory work environment
Qualifications

Education Minimum Requirement:  
  • PhD with less than 3 years of Postdoctoral Experience or achieved a  Ph.D. by Spring/Summer of  2018 
 
Required Experience and Skills:
  • Ability to conceive, design, execute and communicate complex experiments in the field of genetic code expansion, as evidenced by publications
  • Molecular biology and protein expression optimization 
  • Cell imaging, WB, IP
  • Must be an independent and self-motivated scientist
 
Preferred Experience and Skills:
  • Proteostasis experience, signal transduction experience
  • Proteomics experience, bioinformatics experience
  • Optimization of bioorthogonal chemistry for cell-based applications
