The Future Talent Program features internships that last 9-12weeks and the Early Development and Discovery Sciences Department has an internshipwithin Immuno-Oncology Discovery Research in Palo Alto, CA.

T cell populations contain T cells with different antigenspecificities, maturation stage and expression of co-stimulatory orco-inhibitory immune regulatory receptors (IMR).   Therefore it isdifficult to predict T cell responsiveness and outcome of interactions of IMR’s and their ligands at the population level. Establishment of T cell clonesdoes provide an opportunity to study IMR biology in a physiological manner atthe single cell level.  T cell clones have been established againstallo-antigens expressed on antigen presenting cells (APC) such as EBVtransformed B cells and Melanoma cells. These APC have been modified to expressIMR ligands such as PD-L1, CD155 or both.  The characteristics of these Tcells with respect to function (proliferation, cytotoxicity, cytokineproduction) and phenotype as well as responsiveness to IMR manipulation on APCwill be studied in in vitro experiments using a number of cellular immunologytechniques.

The primary duties of this internship will involvecharacterization of human T cell clone function by immune checkpointinhibition. The selected candidate will study the role of immune checkpointinhibitors on human T cell function and ways to increase anti-tumorresponses.  Experience will be obtained in cell isolation, T cell culture,FACS analyses, ELISA and PCR techniques.
