Evaluationof InCell Dx platform for cell sorting/gene expression analysis

Stainingand sorting primary cells for protein and gene expression analysis is highpriority area for Translational Medicine. The current methods aretime-consuming and limited to markers that are expressed on cell surface. Anew technology developed by Incell allows for the fixation, permeabilizationand staining of cells while preserving RNA integrity. The primary advantageof this approach is the ability to sort cells for gene expression analysisbased on both, surface and intracellular proteins. 

Evaluation of the InCell technology was recently prioritized byTranslational Oncology Technology Development Team.  The goal of the summer intern’s project willbe to evaluate the IncellDx platform. First, InCell platform will be compared to traditional flow cytometry.Experiments will be performed using normal and activated human PBMCs andquality of the surface and intracellular staining using IncellDx technologywill be compared to flow cytometry staining. Once comparability is established,cells stained using InCell method will be sorted for gene expression analysis.The quality of RNA will be evaluated against RNA derived from unfixed cellssorted in parallel. Gene expression results obtained using InCell protocol willbe compared to the data obtained from cells sorted using establishedmethod.  At the end of the program, theintern will obtain the key data for the platform assessment and will gainexperience in flow cytometry, cell sorting and gene expression analysis.  PBMC/flow cytometry/cell sorting work will besupervised by Romina Riener.  RNAextraction and gene expression analysis will be supervised by Karen Ku.



  • Candidates muse be currently enrolled in a minimum of a BS/BA in: Biology, Biochemistry, Chemistry or relevant discipline
  • Minimum GPA of 3.0
Required Experience:
  • Candidates must be available for a period of 9-12 continuous weeks, beginning June 2018
